Overview

The sialyl Lewis-keyhole limpet hemocyanin (KLH) conjugate vaccine is an investigational cancer immunotherapy designed to induce an immune response against tumor cells expressing the carbohydrate antigen sialyl Lewis a (CA19-9). The vaccine consists of the oligosaccharide antigen sialyl Lewis a chemically linked to KLH, a nonspecific immunomodulator. Upon administration—typically with the adjuvant QS-21—the vaccine stimulates production of IgG and IgM antibodies targeting the tumor-associated carbohydrate antigen. This immune response may trigger antibody-dependent cell-mediated cytotoxicity (ADCC) against cancer cells expressing the target antigen. Clinical trials have demonstrated that this approach is safe and immunogenic in humans, particularly in patients with metastatic breast cancer[2][3][5][6]. The primary indication under investigation has been breast cancer; however, broader applications in oncology are being explored.
