SiB-2101 is an investigational *in vivo* monocyte immunotherapy being developed by Signadori Bio for the treatment of solid tumors. As the lead candidate in the company's pipeline, it utilizes a proprietary platform designed to engineer monocytes directly within the patient's body to modulate their function and enhance anti-tumor immunity. The therapy specifically targets p21 (encoded by the *CDKN1A* gene) and an additional undisclosed molecular target (Target 1). By modulating p21-mediated pathways, SiB-2101 aims to reprogram the myeloid compartment of the tumor microenvironment, converting immunosuppressive cells into pro-inflammatory effectors that promote a robust anti-cancer immune response. The program is currently in the preclinical stage, with candidate nomination expected in the first quarter of 2027 and the initiation of a first-in-human clinical trial projected for the fourth quarter of 2028.
