SiB-2121 is an experimental *in vivo* monocyte immunotherapy being developed by Signadori Bio for the treatment of solid tumors. It is part of a platform designed to engineer monocytes directly within the body to modulate monocyte-driven anti-tumor immune responses. The therapy specifically targets p21 (encoded by the *CDKN1A* gene) along with an undisclosed third target (Target 3). By modulating p21, the therapy aims to alter the functional state of monocytes and macrophages within the tumor microenvironment to promote an anti-tumor phenotype and overcome immune evasion. As of early 2024, SiB-2121 is in the discovery stage of development.
