siBAHD1 is a small interfering RNA (siRNA) designed to silence the expression of the Bromo Adjacent Homology Domain Containing 1 (BAHD1) protein. BAHD1 is a chromatin-associated factor that plays a critical role in the formation of heterochromatin and the silencing of genes, specifically through the recruitment of histone methyltransferases that catalyze H3K9me3 and H3K27me3 modifications. Research presented at the AACR 2025 Annual Meeting identified BAHD1-dependent heterochromatin formation as a key mechanism underlying acquired radioresistance in prostate and head and neck cancers. By knocking down BAHD1, siBAHD1 reduces the expression of repressive heterochromatin markers and restores sensitivity to radiation in previously resistant cancer cell models. This suggests that targeting the BAHD1-heterochromatin axis may provide a novel therapeutic approach to overcome radioresistance in clinical oncology.
