sibeprelimab

Sibeprelimab is a humanized monoclonal antibody designed to target and neutralize A Proliferation-Inducing Ligand (APRIL), a cytokine belonging to the tumor necrosis factor (TNF) superfamily. APRIL is a critical regulator of B-cell maturation, proliferation, and survival, and its dysregulation is implicated in the production of pathogenic galactose-deficient IgA1 (Gd-IgA1) in IgA nephropathy. By binding to APRIL, sibeprelimab prevents its interaction with its cognate receptors, B-cell maturation antigen (BCMA) and transmembrane activator and CAML interactor (TACI). This blockade leads to a reduction in the levels of pathogenic IgA1 and modulates B-cell-mediated autoimmune responses. Developed by Visterra, a subsidiary of Otsuka Pharmaceutical, sibeprelimab is currently in late-stage clinical development for IgA nephropathy and is also being evaluated for Sjögren’s disease.
