Sibiromycin is a naturally occurring antitumor antibiotic belonging to the pyrrolobenzodiazepine (PBD) class, originally isolated from *Streptosporangium sibiricum*. It functions as a potent DNA minor groove binding agent that forms covalent adducts with DNA, leading to cell death. The compound's structure includes an amino sugar at the C-7 position, which significantly contributes to its high DNA affinity and cytotoxic potency. While historical clinical development of sibiromycin as a standalone systemic therapeutic was limited by cardiotoxicity, it is currently utilized as a cytotoxic payload in the development of antibody-drug conjugates (ADCs).
