SIBP-03 is a recombinant humanized monoclonal antibody that targets the human epidermal growth factor receptor 3 (HER3, also known as ERBB3), a member of the EGFR family implicated in tumor cell survival and proliferation. By binding to HER3, SIBP-03 inhibits both neuregulin-dependent and -independent activation of HER3, thereby blocking downstream signaling pathways such as PI3K/AKT that are crucial for cancer cell growth. The drug exerts antitumor effects through direct inhibition of HER3-mediated signaling and by inducing immune-mediated mechanisms including antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Preclinical studies have shown that SIBP-03 enhances the efficacy of EGFR-targeted (cetuximab) or HER2-targeted (trastuzumab) therapies when used in combination. It is currently being developed for advanced malignant solid tumors, including recurrent head and neck squamous cell carcinoma, esophageal squamous cell carcinoma, and breast cancer with high HER2 expression[1][2][4][6].
