Overview

Sibrotuzumab is a humanized monoclonal antibody that targets fibroblast activation protein alpha (FAP), a cell surface serine protease highly expressed on activated stromal fibroblasts in the tumor microenvironment of many epithelial cancers. By binding to FAP, sibrotuzumab acts as an antagonist and is designed to disrupt the supportive tumor stroma and potentially inhibit tumor growth. It was developed primarily for the treatment of various malignancies including colorectal cancer, non-small cell lung cancer, breast cancer, pancreatic cancer, and head and neck cancers. Despite promising preclinical rationale and early-phase clinical trials demonstrating safety in patients with advanced FAP-positive cancers, clinical development was discontinued after failing to demonstrate efficacy in phase II trials for metastatic colorectal cancer[1][2][5][6].
