Sibrotuzumab-IR700DX is an antibody-photosensitizer conjugate (APC) designed for near-infrared photoimmunotherapy (NIR-PIT). It consists of the humanized monoclonal antibody sibrotuzumab, which targets fibroblast activation protein (FAP), conjugated to the photosensitizer IRDye700DX. This therapeutic agent is designed to selectively target and eliminate FAP-expressing cancer-associated fibroblasts (CAFs) within the tumor microenvironment. Upon exposure to near-infrared light, the IR700DX moiety induces rapid, selective cell death of the targeted CAFs, which can lead to the remodeling of the extracellular matrix and improved delivery of co-administered chemotherapeutic agents. Preclinical studies in esophageal cancer models have demonstrated its potential to overcome drug resistance by reducing interstitial pressure and enhancing drug penetration.
