siCAMK1D is a small interfering RNA (siRNA) designed to specifically target and silence the expression of Calcium/calmodulin-dependent protein kinase ID (CAMK1D). CAMK1D is a kinase involved in various signaling pathways, and its overexpression has been linked to the maintenance of prostate cancer stem-like cells (PCaSCs) and the promotion of an immunosuppressive tumor microenvironment in enzalutamide-resistant prostate cancer. By utilizing RNA interference (RNAi), siCAMK1D reduces CAMK1D protein levels, which in turn suppresses the phosphorylation of CREB and downregulates key stemness genes such as CD44 and CD133. This molecular intervention helps to reverse enzalutamide resistance, decrease regulatory T cell infiltration, and enhance CD8+ T cell-mediated anti-tumor immunity. It is often studied in conjunction with targeted delivery systems, such as CD44-targeted nanoparticles (e.g., EC@HNA), to improve its therapeutic index and restore tumor-immune surveillance.
