siEphA2-LNP is an experimental lipid nanoparticle (LNP) formulation encapsulating small interfering RNA (siRNA) designed to target and silence the expression of Ephrin type-A receptor 2 (EphA2). EphA2 is a receptor tyrosine kinase that is frequently overexpressed in various cancers, including breast and prostate cancers, and plays a critical role in tumor progression, metastasis, and therapy resistance. Developed by researchers at the Houston Methodist Research Institute, Texas A&M University, and the University of Louisville, siEphA2-LNP is being investigated for the treatment of breast cancer liver metastases (BCLM) and triple-negative breast cancer (TNBC). By delivering EphA2-targeting siRNA, the formulation downregulates EphA2 expression, which modulates the tumor microenvironment by polarizing tumor-associated macrophages from an immunosuppressive M2 phenotype to a pro-inflammatory, anti-tumorigenic M1 phenotype, thereby reducing tumor burden and enhancing therapeutic efficacy.
