Overview

siEPHB4 is a small interfering RNA (siRNA) designed to silence the expression of the Ephrin type-B receptor 4 (EPHB4). EPHB4 is a receptor tyrosine kinase that is frequently overexpressed in various malignancies, including prostate, breast, and lung cancers, where it promotes tumor growth, survival, and angiogenesis. By mediating the cleavage of EPHB4 mRNA through the RNA-induced silencing complex (RISC), siEPHB4 reduces EPHB4 protein levels. This downregulation leads to the inhibition of oncogenic signaling (such as the SRC/p38 MAPK/4EBP1 cascade), activation of endoplasmic reticulum (ER) stress, and induction of immunogenic cell death (ICD). In prostate cancer models, siEPHB4 has been shown to decrease GLUT3 expression and ATP levels, further contributing to its anti-tumor effects by impairing glucose metabolism.
