Drug intelligence / Profile preview
siERBB3 is a small interfering RNA (siRNA) designed to target and silence the expression of receptor tyrosine-protein kinase erbB-3 (ERBB3/HER3). Developed by researchers at the University of Pittsburgh, siERBB3 is formulated within lipid nanoparticles (LNPs) for in vivo delivery. In preclinical models of metabolic dysfunction-associated steatotic liver disease (MASLD) and metabolic dysfunction-associated steatohepatitis (MASH), therapeutic silencing of hepatocyte ERBB3 via LNP-delivered siERBB3 has been shown to reverse established steatosis and fibrosis. Mechanistically, ERBB3 knockdown attenuates downstream PI3K-AKT-mTOR signaling, suppresses PPAR-gamma, and restores HNF4-alpha expression, thereby reprogramming the metabolic PPAR-gamma-HNF4-alpha axis to restore hepatic homeostasis and induce lipophagy.
