siFZD2 is a small interfering RNA (siRNA) designed to target and silence the expression of Frizzled-2 (FZD2), a G protein-coupled receptor that serves as a key mediator of non-canonical Wnt5a signaling. In advanced prostate cancer, particularly enzalutamide-resistant castration-resistant prostate cancer (CRPC), the Wnt5a/FZD2 axis is frequently co-upregulated and is associated with tumor progression, neural lineage plasticity, and neuroendocrine differentiation. By knocking down FZD2 expression, siFZD2 inhibits the non-canonical Wnt signaling pathway, suppresses cancer cell proliferation and migration, and resensitizes resistant cells to enzalutamide treatment. Research conducted at the University of California Davis identifies FZD2 as a critical driver of antiandrogen resistance, suggesting that its inhibition could provide therapeutic benefits for patients with advanced prostate cancer expressing high levels of lineage markers.
