SIG-001 is an encapsulated allogeneic cell therapy developed by Sigilon Therapeutics (acquired by Eli Lilly) for the treatment of severe or moderately-severe hemophilia A. The therapy utilizes Sigilon's Shielded Living Therapeutics (SLTx) platform, which consists of engineered human cells encapsulated within a proprietary Afibromer matrix. This matrix is designed to protect the cells from the host immune system and prevent the foreign body response (fibrosis), thereby eliminating the need for systemic immunosuppression. The encapsulated cells are genetically modified with a non-viral vector to continuously produce and secrete B-domain deleted human Factor VIII (BDD-hFVIII). SIG-001 is administered laparoscopically into the peritoneal cavity.
