sig-HELP-E7SH-kdel is an investigational therapeutic DNA vaccine targeting the HPV16 E7 oncoprotein, developed by the Netherlands Cancer Institute (NKI-AVL). The vaccine is a plasmid-based construct (pUMVC3 backbone) encoding a shuffled (rearranged) HPV16 E7 sequence (E7SH) to prevent oncogenic transformation while maintaining immunogenicity. The construct is engineered with a signal peptide ('sig') for endoplasmic reticulum (ER) import, a KDEL retention signal to improve antigen processing and presentation by dendritic cells, and CD4 T-helper epitopes (HELP) to enhance the T-cell response. Administered via DNA tattoo vaccination, it has been evaluated in Phase I/II clinical trials for the treatment of HPV16-positive usual vulvar intraepithelial neoplasia (uVIN).
