**siG12D-LODER + folfirinox** is a combination therapy consisting of siG12D-LODER, a miniature biodegradable polymeric implant delivering a small interfering RNA (siRNA) targeting the KRAS G12D mutation, and FOLFIRINOX, a multi-agent chemotherapy regimen (folinic acid, fluorouracil, irinotecan, and oxaliplatin). siG12D-LODER is implanted directly into the tumor where it slowly releases siG12D siRNA, effectively silencing mutant KRAS mRNA expression and inhibiting tumor growth. Combined with FOLFIRINOX, this regimen is under clinical investigation as a targeted first-line treatment for locally advanced, unresectable pancreatic cancer. The siRNA therapy is designed for local, sustained delivery to overcome systemic delivery barriers, displaying anti-tumor, anti-metastatic, and anti-proliferative activity.
