Overview

Siglec-10 blocking antibody refers to a class of monoclonal antibodies designed to inhibit the Sialic acid-binding Ig-like lectin 10 (Siglec-10) receptor. Siglec-10 is an inhibitory immune checkpoint receptor predominantly expressed on tumor-associated macrophages (TAMs). It interacts with sialylated ligands on cancer cells, such as CD24 or Integrin alpha-3 (ITGA3), to transmit a "don't eat me" signal that suppresses macrophage-mediated phagocytosis. By blocking this interaction, these antibodies restore the ability of macrophages to clear tumor cells. Research presented at AACR 2025 by investigators from Northwestern University and The Wistar Institute highlighted the role of the Siglec-10/ITGA3 axis in pancreatic ductal adenocarcinoma (PDAC) immune evasion and demonstrated that Siglec-10 blockade can significantly reduce tumor growth in preclinical models.
