Siglec-6 BBz CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Siglec-6 (sialic acid-binding immunoglobulin-like lectin 6), a protein expressed on the surface of acute myeloid leukemia (AML) blasts, AML stem cells, and chronic lymphocytic leukemia (CLL) cells. The CAR construct incorporates a human monoclonal antibody-derived targeting domain (JML-1), a 4-1BB (CD137) costimulatory domain, and a CD3-zeta signaling domain. A key advantage of this therapy is that Siglec-6 is not expressed on normal hematopoietic stem and progenitor cells (HSPCs), potentially allowing for the eradication of leukemia without the severe myelosuppression associated with other AML targets like CD33 or CD123. Preclinical data have demonstrated potent cytolytic activity and the ability to induce complete remission in xenograft models.
