Siglec-6 CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target sialic acid-binding immunoglobulin-like lectin 6 (Siglec-6, also known as CD327). Siglec-6 is a cell-surface protein that is highly and commonly expressed on leukemic blasts and leukemic stem cells in acute myeloid leukemia (AML), as well as on transformed B cells in chronic lymphocytic leukemia (CLL). A distinguishing feature of Siglec-6 as a therapeutic target is its absence on normal hematopoietic stem and progenitor cells (HSPCs), which contrasts with other common AML targets like CD33 or CD123. This profile suggests that Siglec-6 CAR T-cell therapy could potentially eliminate malignant cells without causing long-term bone marrow toxicity or requiring subsequent hematopoietic stem cell transplantation. The CAR construct typically utilizes a targeting domain derived from the human monoclonal antibody JML-1 to direct T-cell mediated cytotoxicity against Siglec-6-expressing tumor cells.
