Drug intelligence / Profile preview
Siglec-9 monoclonal antibodies are immunomodulatory agents designed to target Sialic acid-binding Ig-like lectin 9 (Siglec-9), an inhibitory receptor predominantly expressed on immune cells of myeloid and lymphoid lineages, including neutrophils, monocytes, macrophages, dendritic cells, and natural killer (NK) cells. Depending on their functional properties, these antibodies can act as agonists or antagonists. Agonistic Siglec-9 antibodies mimic natural sialic acid ligands to trigger inhibitory ITIM-mediated signaling, which can suppress inflammatory responses, reduce reactive oxygen species (ROS) production, and induce apoptosis in neutrophils, offering potential therapeutic avenues for inflammatory and autoimmune diseases. Conversely, antagonistic (blocking) Siglec-9 antibodies disrupt the interaction between Siglec-9 and its sialoglycan ligands overexpressed on cancer cells, thereby reversing immune suppression and enhancing anti-tumor immunity, making them attractive candidates for cancer immunotherapy.
