Siglec6 CAR-T is an investigational chimeric antigen receptor T-cell (CAR-T) therapy that targets sialic acid–binding immunoglobulin-like lectin 6 (Siglec-6), a cell surface antigen highly expressed on acute myeloid leukemia (AML) cells and a subset of chronic lymphocytic leukemia (CLL) cells. The CAR uses a targeting domain derived from the JML-1 human monoclonal antibody. Preclinical studies show that Siglec6 CAR-T cells specifically eliminate AML blasts (including leukemic stem cells) and CLL cells, induce remission in AML xenograft models, and spare normal hematopoietic stem and progenitor cells (HSPCs), which do not express Siglec-6. This specificity offers the potential for effective leukemia clearance with a lower risk of myeloablation compared to other myeloid-targeted CAR-T therapies. Limited on-target off-tumor reactivity may occur, as Siglec-6 is also present on some mature B cells and basophils[1][2][5][8].
