**SigmE7 + MtHSP70 DNA vaccine** is an experimental therapeutic DNA vaccine developed for the treatment of HPV-related cancers, particularly cervical cancer. The vaccine is constructed by fusing a signal peptide gene sequence from human CD33 (Sig) upstream to a chimeric gene containing a modified human papillomavirus type 16 E7 antigen (mE7) and the heat shock protein 70 gene from Mycobacterium tuberculosis (MtHSP70). The primary mechanism involves the induction of strong HPV16 E7-specific CD8+ T cell immune responses, with the fusion protein acting both as a tumor antigen (E7) and an immunostimulatory adjuvant (HSP70), thereby enhancing cytotoxic T lymphocyte activation and antitumor effects in preclinical mouse models[1][2][3]. The addition of the CD33 signal sequence aims to further direct the fusion protein for secretion and antigen presentation.
