**siHSP47-NP** is a preclinical, intradermally administered antifibrotic nanoparticle therapy developed by PDX Pharmaceuticals for scleroderma-associated skin fibrosis. It uses PDX-NP, a mesoporous silica nanoparticle with polyethylenimine and polyethylene glycol coatings, to deliver a small interfering RNA directed against heat shock protein 47. HSP47 silencing is intended to reduce collagen processing and fibrotic matrix accumulation, while the antioxidant silica core scavenges reactive oxygen species and is associated with reduced NADPH oxidase 4 and other profibrotic markers in preclinical models.
