SIINFEKL-P30 is a chimeric peptide vaccine construct used primarily as a model in immunological research to study the enhancement of CD8+ T-cell responses. It consists of the MHC class I-restricted epitope SIINFEKL (derived from chicken ovalbumin, residues 257–264) fused or conjugated to the P30 universal T-helper epitope (derived from tetanus toxin, residues 947–967). The P30 sequence provides potent CD4+ T-cell help, which is critical for the induction and maintenance of robust SIINFEKL-specific cytotoxic T-lymphocyte (CTL) responses. This construct is frequently employed in preclinical studies to evaluate novel vaccine delivery platforms, adjuvants, and the mechanisms of T-cell activation and memory formation in murine models, specifically those expressing the H-2Kb MHC molecule.
