Overview

SIK2 inhibitor refers to a class of small-molecule therapeutics, specifically a first-in-class selective program developed by Nimbus Therapeutics, designed to target salt-inducible kinase 2 (SIK2) for the treatment of inflammatory bowel disease (IBD). SIK2 is a member of the AMPK-related kinase family that regulates the production of inflammatory cytokines in myeloid cells. By selectively inhibiting SIK2, these compounds suppress the production of pro-inflammatory cytokines such as TNFα, IL-12/23, and IL-6, while simultaneously promoting the release of anti-inflammatory IL-10 and tissue repair factors like amphiregulin (AREG). Preclinical studies in colitis models demonstrate that selective SIK2 inhibition reduces colonic damage and accelerates mucosal healing by upregulating barrier integrity markers, including mucin-2, occludin, and zonula occludens-1 (ZO-1). This mechanism represents a novel oral therapeutic approach for managing chronic intestinal inflammation and promoting regeneration.
