siKDM6A is a small interfering RNA (siRNA) designed to silence the expression of the histone demethylase KDM6A (also known as UTX). In preclinical studies, particularly in glioblastoma stem-like cells (GSCs) and glioma cell lines, siKDM6A has been used to investigate the role of KDM6A in radiosensitivity. By knocking down KDM6A, the radiation-induced decrease of histone H3K27 tri-methylation (H3K27me3) is abolished, leading to a significant delay in the dispersal of γH2AX nuclear foci. This suggests that KDM6A inhibition or knockdown impairs DNA double-strand break (DSB) repair, thereby sensitizing glioblastoma cells to radiotherapy.
