Overview

sIL-6R antibody refers to a class of monoclonal antibodies designed to specifically target the soluble form of the interleukin-6 receptor (sIL-6R). Interleukin-6 (IL-6) signaling can occur through two distinct pathways: classic signaling, which involves the membrane-bound IL-6 receptor (mIL-6R), and trans-signaling, which involves the soluble receptor. While classic signaling is often associated with regenerative and anti-inflammatory functions, trans-signaling is primarily linked to pro-inflammatory and pro-tumorigenic activities. By binding to sIL-6R, these antibodies prevent the formation of the IL-6/sIL-6R complex, thereby blocking its ability to activate the gp130 signal transducer on cells that do not express mIL-6R. In the context of pancreatic cancer research, sIL-6R antibodies have been shown to abrogate mesothelin-induced cell survival and proliferation by disrupting this autocrine/paracrine trans-signaling axis.
