siLuc-LODER is an experimental RNA interference (RNAi) formulation consisting of a small interfering RNA (siRNA) targeting the luciferase gene, delivered via the LODER™ (Local Delivery EluteR) platform. Developed by Silenseed Ltd., the LODER™ is a miniature, biodegradable polymeric scaffold designed for intratumoral implantation. It provides sustained release of the siRNA payload over several months, aiming to achieve long-term gene silencing within the tumor microenvironment. In preclinical and clinical research, siLuc-LODER is primarily utilized as a non-therapeutic negative control to evaluate the sequence-specific efficacy of other siRNA-LODER products, such as siG12D-LODER, which targets mutated KRAS in pancreatic cancer. The delivery system is engineered to overcome physical barriers in solid tumors by utilizing a multi-step distribution process involving priming, convection, and diffusion.
