silver(dppe)2 triflate is an investigational silver(I) coordination complex featuring two 1,2-bis(diphenylphosphino)ethane (dppe) ligands and a triflate counterion. This compound has demonstrated significant antineoplastic activity, particularly in models of drug-resistant ovarian cancer such as MES-OV cells. Unlike conventional platinum-based therapies or many other silver complexes that target thioredoxin reductase or induce apoptosis, silver(dppe)2 triflate triggers paraptosis. Paraptosis is a form of programmed cell death characterized by extensive cytoplasmic vacuolization, often originating from the swelling of the endoplasmic reticulum and mitochondria. Research indicates that silver(dppe)2 triflate is effective even in p53-deficient or platinum-resistant cell lines, suggesting its potential as a candidate for treating refractory malignancies.
