Drug intelligence / Profile preview
Simón bacteriophage (øSimón) is an environmental siphovirus bacteriophage developed by Monash University and Alfred Health for the treatment of multidrug-resistant *Bordetella bronchialis* infections. It was isolated from wastewater in Melbourne, Australia, and characterized as a siphovirus with a 60,921 bp genome. øSimón targets *B. bronchialis* by binding to the outer membrane cobalamin transporter BtuB. It has been evaluated under compassionate use in a patient with cystic fibrosis suffering from recurrent, life-threatening *B. bronchialis* bacteremia and pneumonia. However, the therapy failed due to pre-existing neutralizing antibodies that cross-reacted with the therapeutic phage, driven by the patient's immune response to induced prophages from the causative bacteria, as well as bacterial heteroresistance.
