SIM0226 is an investigational antibody-drug conjugate (ADC) developed by Simcere Zaiming (the oncology subsidiary of Simcere Pharmaceutical) for the treatment of advanced solid tumors. The drug consists of a humanized monoclonal antibody targeting Protein Tyrosine Kinase 7 (PTK7) conjugated to a potent cytotoxic payload, typically a topoisomerase I inhibitor, via a cathepsin-cleavable linker. PTK7, also known as colon carcinoma kinase 4 (CCK4), is a pseudo-kinase receptor overexpressed in various malignancies, including non-small cell lung cancer (NSCLC), triple-negative breast cancer (TNBC), and esophageal cancer, while having limited expression in normal tissues. Upon binding to PTK7 on the cell surface, SIM0226 is internalized, and the payload is released to induce DNA damage and apoptosis. It is currently being evaluated in Phase 1 clinical trials to assess its safety, tolerability, and preliminary efficacy.
