SIM0236 (also known as MG-021) is a bispecific monoclonal antibody targeting both Programmed Death-Ligand 1 (PD-L1) and Roundabout Homolog 1 (ROBO1). Developed by Medgenn Bio (Shandong Simcere Medgenn), a subsidiary of Simcere Pharmaceutical, this therapeutic agent is designed to simultaneously block the PD-1/PD-L1 immunosuppressive checkpoint and inhibit ROBO1-mediated tumor signaling pathways. ROBO1 is a transmembrane receptor that, upon binding its ligand SLIT2, promotes tumor cell migration, invasion, and angiogenesis in various solid tumors. By dual-targeting these pathways, SIM0236 aims to restore anti-tumor immune activity while directly suppressing tumor progression and metastasis. The drug is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors.
