SIM0237 is a bi-functional fusion protein designed for cancer immunotherapy. It consists of a monoclonal antibody targeting programmed cell death ligand 1 (PD-L1) fused with a potency-reduced interleukin-15 (IL-15)/IL-15 receptor alpha (IL-15Rα) sushi domain complex. The drug blocks the PD-1/PD-L1 immunosuppressive pathway by binding to PD-L1, thereby relieving tumor-induced immune suppression. Simultaneously, the IL-15 component activates and expands natural killer (NK) cells and CD8+ T cells, boosting antitumor immune responses. The attenuation of IL-15 activity increases the maximum tolerated dose and reduces risks of overstimulation or anergy in immune cells. Fusion with anti-PD-L1 also enhances the half-life of IL-15 and targets its delivery to the tumor microenvironment, minimizing systemic toxicity such as cytokine release syndrome. SIM0237 is being developed primarily for advanced solid tumors and non-muscle invasive bladder cancer[1][2][3][4][5].
