SIM0270 is a brain-penetrant, oral, small molecule selective estrogen receptor degrader (SERD) developed for the treatment of estrogen receptor-positive (ER+), human epidermal growth factor receptor 2-negative (HER2-) locally advanced or metastatic breast cancer. It acts by antagonizing and degrading the estrogen receptor alpha (ERα), thereby inhibiting ER-driven tumor growth and proliferation. SIM0270 has demonstrated favorable safety and early antitumor activity in clinical trials, including in patients with ESR1 mutations who have progressed on prior endocrine therapies. The drug is being evaluated both as monotherapy and in combination with agents such as everolimus or palbociclib to address resistance to current endocrine treatments[1][3][5][7]. SIM0270 was independently developed by Simcere Zaiming, a subsidiary of Simcere Pharmaceutical Group.
