Overview

SIM0270 + everolimus is an investigational oral combination therapy for the treatment of estrogen receptor-positive, HER2-negative (ER+/HER2-) locally advanced or metastatic breast cancer. SIM0270 is a novel, brain-penetrant, oral selective estrogen receptor degrader (SERD) designed to degrade the estrogen receptor and overcome resistance to prior endocrine therapies. Everolimus is a well-established mammalian target of rapamycin (mTOR) inhibitor that disrupts cellular growth and proliferation pathways. The combination aims to leverage potential synergistic effects by targeting both ER signaling and mTOR pathways, addressing limited efficacy seen with current endocrine therapies in patients who have progressed after CDK4/6 inhibitors or other standard treatments[1][3][4][5][7].
