Overview

SIM0323 is a humanized IgG1 monoclonal antibody designed to target the V-domain Ig suppressor of T cell activation (VISTA), an immune checkpoint protein. Developed by Simcere Pharmaceutical (specifically through its subsidiary Shandong Simcere Medgenn), SIM0323 acts as a VISTA antagonist, blocking the inhibitory signaling that VISTA exerts on T cells and myeloid cells within the tumor microenvironment. By neutralizing this checkpoint, the drug aims to restore and enhance anti-tumor immune responses. SIM0323 is primarily being developed for the treatment of advanced solid tumors, particularly in patients who may be refractory to existing PD-1/PD-L1 therapies. It is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and preliminary efficacy.
