SIM0327 is an investigational multi-specific antibody developed by Simcere Pharmaceutical (Jiangsu Xiansheng). It is designed to simultaneously target Programmed Death-Ligand 1 (PD-L1) and 4-1BB (CD137), a co-stimulatory receptor on T cells. By blocking the PD-1/PD-L1 inhibitory pathway, SIM0327 aims to release the immune system's brakes, while its 4-1BB agonistic activity provides a localized co-stimulatory signal to enhance T-cell proliferation and anti-tumor activity. This dual-targeting approach is intended to improve therapeutic efficacy in oncology while potentially minimizing the systemic toxicities often associated with traditional 4-1BB agonists. The drug is currently in early-stage development for the treatment of various cancers.
