SIM0348 is a humanized IgG1-based bispecific antibody that targets both TIGIT (T cell immunoreceptor with Ig and ITIM domains) and PVRIG (Poliovirus receptor-related immunoglobulin domain-containing protein). It is designed to enhance anti-tumor immune responses by simultaneously blocking these two inhibitory receptors on immune cells. The drug activates cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, thereby promoting immune-mediated tumor cell killing and inhibiting tumor proliferation. Additionally, SIM0348 induces Fc-mediated depletion of immunosuppressive regulatory T cells (Tregs) expressing high levels of TIGIT or both TIGIT and PVRIG. Preclinical studies have shown that it enhances NK cell-mediated killing of cancer cells and increases IFN-γ secretion by antigen-specific CD8+ T cells more effectively than monotherapies targeting either receptor alone. Developed for the treatment of advanced malignancies, SIM0348 has demonstrated synergistic effects when combined with anti-PD-L1 antibodies in preclinical models[1][3][4][5][7].
