SIM0348 + sintilimab is an experimental combination therapy under investigation for the treatment of advanced malignancies, particularly advanced solid tumors. SIM0348 is a humanized IgG1-based bi-specific monoclonal antibody developed by Simcere that targets both TIGIT and PVRIG, two immune checkpoint proteins. It blocks the interactions of CD155/TIGIT and CD112/PVRIG, aiming to release immune suppression and enhance anti-tumor immunity by promoting NK cell and antigen-specific CD8+ T cell activity, as well as depleting immunosuppressive Treg cells expressing TIGIT and PVRIG. Sintilimab, marketed as Tyvyt, is a humanized IgG4 monoclonal antibody targeting PD-1, developed by Innovent and Eli Lilly, and functions as a checkpoint inhibitor by blocking PD-1/PD-L1 signaling to activate T-cells against tumors. The combination of SIM0348 with other checkpoint inhibitors such as anti-PD-L1 has shown synergistic antitumor activity in preclinical studies. Early-phase clinical trials for SIM0348 in advanced solid tumors have recently begun, and clinical studies on checkpoint inhibitor combinations (including sintilimab) are a focus of current immuno-oncology research[1][2][4].
