SIM0366 is a small molecule inhibitor of the Werner syndrome ATP-dependent helicase (WRN), currently in development by Simcere Pharmaceutical (specifically its subsidiary Shandong Simcere Medgenn) for the treatment of oncology. WRN is a member of the RecQ helicase family and has been identified as a key synthetic lethal target in cancers characterized by high microsatellite instability (MSI-H) or deficient DNA mismatch repair (dMMR). By inhibiting WRN helicase activity, SIM0366 induces DNA damage and cell death selectively in MSI-H/dMMR tumor cells while sparing normal cells, offering a targeted therapeutic approach for patients with these specific genetic alterations. The drug is currently in the pre-clinical/IND-enabling stage of development.
