SIM0500 is a humanized trispecific antibody designed as a T-cell engager for the treatment of relapsed or refractory multiple myeloma. Developed by Simcere Zaiming using their proprietary polyspecific antibody technology platform, it targets three antigens: GPRC5D (G-protein coupled receptor family C group 5 member D), BCMA (B-cell maturation antigen), and CD3 on T cells. The molecule consists of an IgG4-based structure with a CD3-specific single-chain Fv flanked by anti-BCMA and anti-GPRC5D antibodies. By binding to both tumor antigens (BCMA and/or GPRC5D) on multiple myeloma cells and CD3 on T-cells, SIM0500 recruits activated T-cells to kill malignant plasma cells expressing these markers. Preclinical studies have shown strong tumor-killing effects and good tolerability at low doses. The drug has received Fast Track designation from the FDA for patients with relapsed or refractory multiple myeloma who have failed established therapies[1][4][6][8].
