Overview

SIM0520 is an investigational small molecule inhibitor of DNA polymerase theta (Polθ), developed by Simcere Pharmaceutical and its subsidiary Shandong Simcere Medgenn. Polθ is a specialized DNA polymerase that plays a critical role in the microhomology-mediated end joining (MMEJ) pathway, a DNA repair mechanism that becomes essential in cancer cells with homologous recombination deficiency (HRD), such as those harboring BRCA1 or BRCA2 mutations. By targeting Polθ, SIM0520 aims to exploit synthetic lethality, selectively killing HR-deficient tumor cells while sparing healthy tissue. The drug is currently in preclinical development for the treatment of solid tumors, potentially as a monotherapy or in combination with PARP inhibitors or other DNA-damaging agents.
