SIM0596 is an orally bioavailable, small molecule dual inhibitor of phosphoinositide 3-kinase (PI3K) and the mammalian target of rapamycin (mTOR), currently under development by Simcere Pharmaceutical. The PI3K/AKT/mTOR pathway is a critical signaling axis that regulates cell growth, proliferation, and survival, and its dysregulation is a hallmark of many human cancers. By simultaneously targeting both PI3K and mTOR, SIM0596 is designed to provide a more comprehensive blockade of this pathway compared to selective inhibitors, potentially overcoming compensatory feedback loops that often lead to therapeutic resistance.
