Overview

SIM0596 is an orally bioavailable, small molecule dual inhibitor of phosphoinositide 3-kinase (PI3K) and the mammalian target of rapamycin (mTOR), currently under development by Simcere Pharmaceutical. The PI3K/AKT/mTOR pathway is a critical signaling axis that regulates cell growth, proliferation, and survival, and its dysregulation is a hallmark of many human cancers. By simultaneously targeting both PI3K and mTOR, SIM0596 is designed to provide a more comprehensive blockade of this pathway compared to selective inhibitors, potentially overcoming compensatory feedback loops that often lead to therapeutic resistance. The drug is primarily being investigated for the treatment of advanced solid tumors.
