SIM0680 is an investigational small molecule anti-tumor agent developed by Simcere Pharmaceutical (Jiangsu Xiansheng Re Ming Pharmaceutical). It is characterized as a "Pan-RB" modulator, a novel therapeutic approach designed to maintain the active, tumor-suppressive state of the Retinoblastoma (RB) protein family, including RB1, RBL1 (p107), and RBL2 (p130). This is achieved through the potent inhibition of multiple cyclin-dependent kinases (CDK2, CDK4, and CDK6) that typically phosphorylate and inactivate RB proteins. By preventing this inactivation, SIM0680 promotes G1 cell cycle arrest and inhibits the proliferation of cancer cells, particularly in tumors that have developed resistance to selective CDK4/6 inhibitors. The drug is currently in the early stages of development for the treatment of various solid tumors.
