SIM0721 is an orally bioavailable small molecule inhibitor of Complement Factor B (CFB), developed by Shandong Simcere Medgenn, a subsidiary of Simcere Pharmaceutical. It specifically targets the alternative pathway of the complement system, which is a key driver of inflammation and tissue damage in various autoimmune and inflammatory disorders. By inhibiting Factor B, SIM0721 prevents the formation of the C3 convertase (C3bBb), thereby blocking the amplification loop of the complement cascade and reducing the production of pro-inflammatory mediators such as C3a, C5a, and the membrane attack complex (MAC). The drug is primarily being developed for the treatment of IgA nephropathy (IgAN) and other complement-mediated diseases, aiming to provide a targeted therapeutic option that mitigates renal damage and systemic inflammation.
