SIM0757 is an antibody-drug conjugate (ADC) targeting the interleukin-4 receptor alpha (IL-4Rα), currently under development by Simcere Pharmaceutical (specifically its subsidiary Shandong Simcere Medgenn). IL-4Rα is a critical subunit of both Type I and Type II IL-4 receptors, mediating the signaling of IL-4 and IL-13, which are key drivers of Type 2 inflammation. While traditional therapies targeting this pathway, such as dupilumab, are monoclonal antibodies that block receptor signaling, SIM0757 utilizes the ADC modality to potentially provide enhanced efficacy through targeted delivery of a cytotoxic or immunomodulatory payload to IL-4Rα-expressing cells. It is primarily being investigated for the treatment of autoimmune and inflammatory diseases.
