SIM0760 is an innovative, recombinant interleukin-2 (IL-2) mutant Fc fusion protein developed by Simcere Pharmaceutical for the treatment of autoimmune diseases. It is engineered as a "Treg-biased" IL-2 agonist, designed to selectively activate and expand regulatory T cells (Tregs) while minimizing the stimulation of pro-inflammatory effector T cells and natural killer (NK) cells. This selectivity is achieved through specific mutations in the IL-2 sequence that enhance its affinity for the high-affinity trimeric IL-2 receptor (comprising CD25, CD122, and CD132) predominantly expressed on Tregs, while reducing its binding to the dimeric IL-2 receptor (CD122 and CD132) found on other immune cells. By promoting Treg-mediated immune tolerance, SIM0760 aims to restore immune homeostasis in patients with conditions such as systemic lupus erythematosus (SLE).
