Overview

SIM0811 is a recombinant humanized thrombolytic protein developed by **Simcere** for the treatment of thrombotic diseases, including acute ischemic stroke. It is a modified version of staphylokinase, a bacterial protein that acts as a potent plasminogen activator. SIM0811 is engineered to have reduced immunogenicity compared to wild-type staphylokinase while maintaining high fibrin selectivity, which minimizes systemic fibrinogen depletion and reduces the risk of bleeding complications. The drug works by forming a 1:1 stoichiometric complex with plasminogen; this complex then converts other plasminogen molecules into plasmin, the primary enzyme responsible for the degradation of fibrin clots. SIM0811 is currently being evaluated in Phase I clinical trials to assess its safety, pharmacokinetics, and impact on thrombotic molecular markers such as plasmin-α2 antiplasmin complex (PIC) and fibrin degradation products (FDP).
