This is a combination of two drugs: - **Simeprevir** (also known as TMC435350) is an oral small molecule inhibitor of the hepatitis C virus (HCV) NS3/4A protease. It blocks viral replication by inhibiting the cleavage of HCV polyproteins necessary for viral maturation. Simeprevir is primarily indicated for the treatment of chronic hepatitis C infection in adults with HCV genotype 1 or 4 and has been used in combination regimens with other antivirals[2][5][7][9]. - **Rifampin** is a broad-spectrum antibiotic that inhibits bacterial DNA-dependent RNA polymerase. It is widely used to treat tuberculosis and other bacterial infections. The combination "simeprevir + rifampin" does not represent a marketed fixed-dose product but may be studied together to assess drug-drug interactions or effects on pharmacokinetics. Rifampin is a strong inducer of CYP3A4 enzymes, which can significantly reduce plasma concentrations and efficacy of drugs like simeprevir that are metabolized by CYP3A4[2][5]. This interaction can lead to reduced antiviral activity and potential treatment failure.
